    This is how I do a scrape. Firstly set up your scope,and net the Koi.Leave the koi in the net in the pond and using a plastic plant label or something similar and gently run the scraper along the body of the koi from just behind the gills to the tail then transfer the mucus to the slide before placing the cover slip on.Drip some pond water on the slide as this will help to keep it moist and make finding the parasites easier.Start at a magnification of 100 then work up to 400 you will see most parasites at 1-300 400 can be a bit more tricky. Do not sedate the koi as this can remove some parasites and make other immobile making them harder to see.
